Eastern Cape police are investigating a case of culpable homicide after a truck ploughed into protesters in Middleburg killing two men on Friday morning.
Spokesperson Colonel Sibongile Soci says protesters blocked all entrances to the town and the situation got extremely tense.
She says a 20-year-old man died at the scene whilst a 38-year-old man who was injured, died later in hospital.
Two other people also sustained serious injuries in the incident.
Protesters have been blocking the N10, N9 and R56 since Wednesday.
It is not clear what the protest is about.
Colonel Soci says any person who might have witnessed the incident is asked to come forward.
The truck driver did stop after the accident and was interviewed by police.
She says the situation remains tense and Public Order Police have been deployed to monitor the situation.
Motorists are advised to make use of alternative routes where possible travelling to Colesburg, Steynsburg, Graaff-Reinet and Cradock.
Meanwhile, SA Trucker received information that the protests will resume on Monday morning after the community met on Sunday afternoon.
Truckers are warned to avoid the area and use alternative routes to avoid being targeted.
